摘要: In this study, transcriptome analysis was performed to identify the genes that confer oxygen tolerance and bile salt tolerance in Lactiplantibacillus plantarum AR113, a probiotic bacterium with potential applications in gastrointestinal environments. The plasmid pIB184 was selected as the vector for gene overexpression, with red fluorescent protein (RFP) used as the reporter gene to facilitate the identification and characterization of anaerobic promoters and bile salt-inducible promoters. This system was employed to screen for anaerobic promoters, and the activities of P2441,P1149, P2418 were found to be upregulated by 5.04-fold, 2.61-fold, and 3.03fold, respectively. P2528,P16090,Pshsp1,Pshsp2,Pshsp3 and Pgrosel exhibited increased activity under 0.1 % bile salt conditions, with fold changes of 1.75, 1.41, 1.99, 2.16, 2.10, and 1.59, respectively. Glycocholate and glycochenodeoxycholate were identified as the bile salts that most strongly induced the activity of these six bile salt-inducible promoters. The results of this study provide valuable insights into the regulatory mechanisms underlying bile salt and oxygen stress tolerance in L. plantarum AR113 and may be useful for the development of engineered lactic acid bacteria or functional strains capable of surviving in harsh gastrointestinal conditions.
